چکیده

Although the structure of thiamin (vitamin B,) has been known for over 40 years (Williams, 1936; Grewe, 1936) and its chemical synthesis accomplished soon thereafter (Williams & Cline, 1936; Andersag & Westphal, 1937; Todd & Bergel, 1937), the biosynthetic pathway to thiamin (3) has still not been elucidated completely. It is well established that the pyrimidine (1) and thiazole (2) moieties are biosynthesized independently and are then joined to give thiamin (Scheme 1) (Leder, 1975). However, there is no general agreement concerning the basic

Thiamin biosynthesis: still yielding fascinating biological chemistry.

The present paper describes the biosynthesis of the thiamin thiazole in Bacillus subtilis and Saccharomyces cerevisiae. The two pathways are quite different: in B. subtilis, the thiazole is formed by an oxidative condensation of glycine, deoxy-D-xylulose 5-phosphate and a protein thiocarboxylate, whereas, in S. cerevisiae, the thiazole is assembled from glycine, NAD and Cys205 of the thiazole s...

A cross-kingdom Nudix enzyme that pre-empts damage in thiamin metabolism.

Genes specifying the thiamin monophosphate phosphatase and adenylated thiazole diphosphatase steps in fungal and plant thiamin biosynthesis remain unknown, as do genes for ThDP (thiamin diphosphate) hydrolysis in thiamin metabolism. A distinctive Nudix domain fused to Tnr3 (thiamin diphosphokinase) in Schizosaccharomyces pombe was evaluated as a candidate for these functions. Comparative genomi...

Biosynthesis of the thiazole moiety of thiamin in Escherichia coli: identification of an acyldisulfide-linked protein--protein conjugate that is functionally analogous to the ubiquitin/E1 complex.

A covalently linked protein--protein conjugate between ThiF and ThiS thiocarboxylate was found in a partially purified coexpressed ThiF/ThiS protein mixture by using Fourier transform mass spectrometry. The Cys-184 of ThiF and the C terminus of ThiS thiocarboxylate were identified to be involved in the formation of this complex by using both mutagenesis and chemical modification methods. A comp...

Analysis of Chlamydomonas thiamin metabolism in vivo reveals riboswitch plasticity.

Thiamin (vitamin B1) is an essential micronutrient needed as a cofactor for many central metabolic enzymes. Animals must have thiamin in their diet, whereas bacteria, fungi, and plants can biosynthesize it de novo from the condensation of a thiazole and a pyrimidine moiety. Although the routes to biosynthesize these two heterocycles are not conserved in different organisms, in all cases exogeno...

The structural and biochemical foundations of thiamin biosynthesis.

Thiamin is synthesized by most prokaryotes and by eukaryotes such as yeast and plants. In all cases, the thiazole and pyrimidine moieties are synthesized in separate branches of the pathway and coupled to form thiamin phosphate. A final phosphorylation gives thiamin pyrophosphate, the active form of the cofactor. Over the past decade or so, biochemical and structural studies have elucidated mos...
